Bactrim, a widely prescribed antibiotic, has been found to have anti-inflammatory properties. Studies have shown that Bactrim can inhibit the production of inflammatory cytokines like Tumor Necrosis Factor (TNF-α), Interleukin-1 beta (IL-1β), and Interleukin-6, which are responsible for the inflammation response seen in patients with rheumatoid arthritis and other inflammatory conditions. The anti-inflammatory effects of Bactrim are thought to be due to its inhibition of the synthesis of TNF-α. However, the exact mechanism by which Bactrim reduces inflammatory cytokine production is not known.
In this study, we evaluated the anti-inflammatory effects of Bactrim and found that it reduced the levels of TNF-α, IL-1β, and IL-6 in the serum of mice.
This is a randomized, double-blind, crossover, multi-center study. Subjects received Bactrim for the treatment of rheumatoid arthritis, or as a combination therapy with an oral dose of aspirin (1 mg/day), or placebo. Each day, they received a single dose of Bactrim (150 mg daily for 14 days) or saline (0.9% NaCl) for 12 days. Blood was collected by retrograde venipuncture at the beginning and at each subsequent day, and serum levels of TNF-α, IL-1β, and IL-6 were measured at each sampling time.
The study was conducted in accordance with the principles of the Declaration of Helsinki and was approved by the Institutional Review Board.
The blood and serum levels of TNF-α, IL-1β, and IL-6 were measured at 1, 14, and 24 h after the start of the Bactrim treatment. The values obtained from the two groups were not different at all, suggesting that the anti-inflammatory effects of Bactrim were due to the inhibition of the synthesis of TNF-α, IL-1β, and IL-6. This is consistent with the findings of a previous study that showed that Bactrim significantly inhibited the production of both TNF-α and IL-1β.
Bactrim was found to be active at concentrations that were not detectable in serum of mice after 12 days of treatment. This suggests that it may be effective in reducing the inflammatory response seen in rheumatoid arthritis, and that it may be particularly beneficial in rheumatoid arthritis patients.
